چکیده
New antifolate-resi slant sublines of mouse leukemia L1210 have been developed. Dihydrofolate reductase activities were determined during early stages of increasing resistance to the folate antagonists. A daily dosage level of 0.75 mg amethopterin/kg used to develop subline FR-1 yielded tumor cells with a tenfold increase of enzyme within three serial transplants. When a new selective influence was imposed by the dichloroamethopterin treatment of mice given implants of amethopterin-resistant subline FR-1, an increase to 40 times the activity of the original parent cells occurred. When dichloroamethopterin was used alone for the development of subline FR-3 without any prior selection of resistant cells with amethopterin, partial resistance to the chloro-derivative occurred without any concomitant rise in dihydrofolate reductase. The development of full resistance to dichloroamethopterin resulted in a significant increase of dihydrofolate reductase activity.

Cytogenetics of Mouse Leukemia L1210. I. Association of a Specific Chromosome with Dihydrofolate Reductase Activity in Amethopterin-treated Sublines.
mouse leukemia L1210 treated with and resistant to various chemotherapeutic agents only those 5 which were resistant to amethopterin (Methotrexate) lacked cells con taming a subtelocentric (ST) marker chromosome regu larly present in parental L1210 cells. Thus there was a correlation between amethopterin resistance and a specific chromosomal change. A subsequent study (17) of several of these s...

The disparate cross-resistance behavior of certain amethopterin-resistant Chinese hamster cell lines to the quinazoline antifolate, methasquin, prompted an investigation of the possibility of the existence of more than one species of dihydrofolate reducÃ-asein these cell populations. The dihydrofolate reductase of four amethopterin-resistant Chinese hamster sublines (DC-3F8/A50, 460-fold resist...

Mutation to amethopterin(4-amino-lo-methylpteroylglutamic acid) resistance in Diplococcus pneumoniae has been shown (1) to occur in two distinct and remotely situated regions of the deoxyribonucleic acid. Comparative growth and cross-resistance studies, also reported at that time, suggested that these genetic regions were also biochemically distinguishable. During investigations seeking to iden...

From a single amethopterin-resistant organism, Streptococcus faecium var. durans strain A, two different dihydrofolate reductases have been obtained as essentially homogeneous proteins in good yield. One of the reductases has a similar substrate specificity and turnover number (about 8000 moles per min per mole of enzyme) to the single reductase found in the amethopterin-sensitive strain of S. ...

Preliminary evidence is presented that indicates that the dihydrofolate reductase activity of amethopterin-sensitive Streptococcus faecium var. durans ATCC 8043 is separable into two dihydrofolate reductases, one of which also reduces folate.
